The FDA has approved Trelegy Ellipta (GSK), a fixed-dose
combination of the inhaled corticosteroid (ICS)
fluticasone furoate, the long-acting antimuscarinic
agent (LAMA) umeclidinium, and the long-acting
beta2-agonist (LABA) vilanterol. It is available as a dry
powder inhaler for once-daily maintenance treatment
of COPD and to reduce COPD exacerbations in patients
with a history of exacerbations. Trelegy Ellipta is
the first three-drug inhaler for treatment of COPD to

Two Drugs for Advanced HER2-Positive Breast Cancer (Enhertu and Tukysa) ...
The FDA has approved two new drugs for treatment
of previously-treated unresectable or metastatic
human epidermal growth factor receptor 2 (HER2)-positive breast cancer: fam-trastuzumab deruxtecannxki
(Enhertu – Daiichi-Sankyo/AstraZeneca), an IV
HER2-directed monoclonal antibody linked to the
topoisomerase I inhibitor DXd, and tucatinib (Tukysa – Seagen), an oral tyrosine kinase inhibitor.
Eliglustat (Cerdelga) - An Oral Drug for Gaucher Disease (online only)

The FDA has approved eliglustat (Cerdelga –
Genzyme), an oral glucosylceramide synthase inhibitor,
for treatment of adults with type 1 Gaucher disease.
Eliglustat is metabolized primarily by CYP2D6. Because
patients who are CYP2D6 ultra-rapid metabolizers may
not achieve therapeutic concentrations and a specific
dosage cannot be recommended for indeterminate
metabolizers, the FDA has approved the drug only
for patients who are extensive, intermediate, or poor
metabolizers of CYP2D6.
In Brief: Two Drugs for Soft-Tissue Sarcoma (online only)
The Medical Letter on Drugs and Therapeutics • May 09, 2016 (Issue 1494)
Leporini et al. A comprehensive safety evaluation of trabectedin
and drug-drug interactions of trabectedin ...
The anthracycline doxorubicin with or without the alkylating agent ifosfamide is the standard first-line treatment for advanced soft-tissue sarcomas. The FDA recently approved the minor groove DNA intercalator trabectedin (Yondelis – Janssen) for treatment of unresectable or metastatic liposarcoma or leiomyosarcoma in patients previously treated with an anthracycline. Trabectedin has been available for years in Europe for treatment of advanced soft-tissue sarcoma. The FDA has also approved the microtubule inhibitor eribulin mesylate (Halaven – Eisai), which was approved earlier for...

Rosuvastatin - a New Lipid-lowering Drug
The Medical Letter on Drugs and Therapeutics • Oct 13, 2003 (Issue 1167)
Rosuvastatin - a New Lipid-lowering Drug ...
Rosuvastatin (Crestor - AstraZeneca), an HMG-CoA reductase inhibitor (or "statin"), was recently approved by the FDA for lowering serum cholesterol and triglyceride concentrations and raising HDL cholesterol levels. Rosuvastatin, like other statins, inhibits the enzyme that catalyzes the rate-limiting step in cholesterol synthesis, but it is claimed to be more potent than the others. All of these drugs must be taken indefinitely; if they are discontinued, lipid levels return to baseline.

.............p 190
Some Interactions with Glucose-Lowering Drugs ...................p 191
Comparison Table ...
Diet, exercise, and weight loss can improve glycemic
control, but most patients with type 2 diabetes
eventually require glucose-lowering pharmacotherapy.
An A1C goal of <7% (while minimizing hypoglycemia)
is recommended for most patients to prevent or
reduce the microvascular complications of diabetes
(retinopathy, nephropathy, neuropathy). An A1C target
of <8% may be appropriate for patients who are older,
have comorbid conditions, or are at risk of serious
hypoglycemia-associated adverse events.
