Ceritinib (Zykadia – Novartis), an oral tyrosine
kinase inhibitor, has received accelerated approval
from the FDA for treatment of patients with anaplastic
lymphoma kinase (ALK)-positive metastatic
non-small cell lung cancer (NSCLC) who have
progressed on or are intolerant to crizotinib (Xalkori).
It is the second tyrosine kinase inhibitor to be approved
for ALK-positive metastatic NSCLC; crizotinib was the
first. Translocations of the ALK gene are found in
about 5% of lung cancers; they occur predominantly
in nonsmokers with adenocarcinoma.

Methadone, an opioid agonist that can be taken orally, has been used for maintenance treatment for many years in the USA (DM Novick et al, Drug Alcohol Depend, 33:235, 1993). Last year, the US Food and Drug Administration approved the distribution and use of L-alpha-acetyl-methadol (LAAM; ORLAAM - Bio Development Corporation, McLean, VA), a long-acting congener of methadone.
