C1INH, attenuated androgen, or antifibrinolytic ...
The FDA has approved Haegarda (CSL Bering), a
human plasma-derived C1 esterase inhibitor (C1INH),
for prophylaxis against hereditary angioedema (HAE)
attacks in adolescents and adults. Haegarda is the
fourth C1INH to be approved by the FDA and the
second to be approved for HAE prophylaxis. Unlike
the other C1INHs, Haegarda is self-administered
subcutaneously rather than intravenously.
Addendum: Drospirenone (Slynd) - A New Progestin-Only Oral Contraceptive
The Medical Letter on Drugs and Therapeutics • Mar 23, 2020 (Issue 1594)
that increase potassium levels, such as the anti-androgen aldosterone receptor antagonist spironolactone ...
In our article on Slynd (Med Lett Drugs Ther 2020; 62:18),
the drospirenone-only oral contraceptive, we mentioned that
drospirenone has antiandrogenic activity that could improve
acne and antimineralocorticoid activity that could cause
hyperkalemia. We should have added that concurrent use of
drospirenone with other drugs that increase potassium levels,
such as the anti-androgen aldosterone receptor antagonist
spironolactone (Aldactone, and generics), which is often
used off-label for treatment of acne, can increase the risk of
hyperkalemia.

The oral poly(ADP-ribose) polymerase (PARP)
inhibitor talazoparib (Talzenna – Pfizer) has been
approved by the FDA for use in combination with the
androgen receptor blocker enzalutamide (Xtandi) for
treatment of adults with homologous recombination
repair (HRR) gene-mutated metastatic castration-resistant
prostate cancer (mCRPC). The drug has
been available since 2018 for treatment of deleterious
or suspected deleterious germline BRCA-mutated
(gBRCAm), HER2-negative locally advanced or

Testosterone Patches for Hypogonadism
The Medical Letter on Drugs and Therapeutics • May 24, 1996 (Issue 975)
lifelong androgen replacement to
prevent osteoporosis and maintain normal muscle mass, erythropoiesis ...
Men with primary or secondary hypogonadism require lifelong androgen replacement to prevent osteoporosis and maintain normal muscle mass, erythropoiesis and sexual function (AM Matsumoto, Endocrinol Metab Clin North Am, 23:857, 1994). Until recently, the standard treatment for male hypogonadism has been an intramuscular injection of a long-acting testosterone ester every two to three weeks, which leads to serum testosterone concentrations that are high for a few days, normal for a few days more, and then may be subnormal until the next dose. Two transdermal preparations of testosterone...

of both androgens and estrogens. Plasma
concentrations peak at about 20-30 years of age ...
Dehydroepiandrosterone (DHEA), an endogenous adrenal steroid, is marketed as a dietary supplement in the US. It is widely promoted to reverse the effects of aging (loss of muscle, memory and libido) and has been used by athletes as a substitute for anabolic steroids. DHEA is banned by the International Olympic Committee, National Collegiate Athletic Association, National Football League and other sports organizations, but it was specifically exempted from becoming a controlled substance in the Anabolic Steroid Control Act of 2004.

Takeda), a subcutaneously-administered plasma
kallikrein inhibitor, for prevention of hereditary
angioedema (HAE) attacks in adults and children
≥12 years old. Lanadelumab is the third drug to be
approved for this indication; the human plasmaderived
