antiretroviral agents for treatment of HIV-1 infection. A
combination of emtricitabine and tenofovir disoproxil
fumarate (FTC/TDF; Truvada) has been available
since 2004 for the same indication. Emtricitabine
and TAF are also available in combination with the
non-nucleoside reverse transcriptase inhibitor
(NNRTI) rilpivirine as Odefsey and with the integrase
strand transfer...

The FDA has approved tisagenlecleucel (Kymriah —
Novartis), an individualized, genetically-modified
cellular product, for treatment of relapsed or refractory
B-cell precursor acute lymphoblastic leukemia (ALL)
in patients ≤25 years old. The patient's own T cells
are genetically modified to express chimeric antigen
receptors (CAR) and then reinfused. Kymriah is the
first CAR T-cell immunotherapy to become available in
the US. A CAR T-cell product for B-cell non-Hodgkin's
lymphoma is expected to be approved soon.

of B cells, promoting T-cell activation and proliferation ...
The FDA has approved axicabtagene ciloleucel
(Yescarta – Kite) for treatment of adults with relapsed
or refractory CD19+ large B-cell lymphoma after ≥2
lines of systemic therapy. Yescarta is an individualized
cellular product prepared from the patient's own T cells,
which are genetically modified to express chimeric
antigen receptors (CAR) and then infused back into
the patient. It is the second CAR T-cell immunotherapy
to become available in the US. Tisagenlecleucel
(Kymriah), a CAR T-cell product previously approved
for treatment of relapsed or refractory B-cell...
Encorafenib (Braftovi) for Metastatic Colorectal Cancer

for use with the mitogen-activated kinase (MEK ...
The FDA has approved the oral kinase inhibitor
encorafenib (Braftovi – Pfizer), in combination with
the epidermal growth factor receptor (EGFR) inhibitor
cetuximab (Erbitux), for treatment of adults with
metastatic colorectal cancer (CRC) with a BRAF
V600E mutation. Encorafenib was approved in 2018
for use with the mitogen-activated kinase (MEK)
inhibitor binimetinib (Mektovi) for treatment of
unresectable or metastatic melanoma with a BRAF
V600E or V600K mutation.
Avapritinib (Ayvakit) for GIST
The Medical Letter on Drugs and Therapeutics • Feb 08, 2021 (Issue 1617)

mutations, including D842V, and KIT mutations,
preventing autophosphorylation and activation ...
The FDA has approved the oral tyrosine kinase
inhibitor avapritinib (Ayvakit – Blueprint Medicines)
for treatment of unresectable or metastatic gastrointestinal
stromal tumors (GISTs) harboring a
platelet-derived growth factor receptor alpha
(PDGFRA) D842V or other PDGFRA exon 18 mutation.
Avapritinib is the first drug to be approved for this
indication in the US.
Aducanumab (Aduhelm) for Alzheimer's Disease
The Medical Letter on Drugs and Therapeutics • Jul 12, 2021 (Issue 1628)

of these agents have been shown to stop ...
Aducanumab-avwa (Aduhelm – Biogen/Eisai), an
IV amyloid beta-directed monoclonal antibody,
has received accelerated approval from the FDA
for treatment of Alzheimer's disease. The approval
was based on the surrogate endpoint of reduction in
amyloid beta plaques in the brain. The manufacturer
is required to conduct an additional randomized
controlled trial to establish that reducing amyloid
beta plaques with aducanumab improves clinical
outcomes in patients with Alzheimer's disease.
