treatment of endocrine-resistant, PIK3CA-mutated,
hormone receptor (HR)-positive, human epidermal
growth factor receptor 2 (HER2)-negative, locally
advanced or metastatic breast cancer. This is the first
approval for Itovebi; palbociclib and fulvestrant have
been available for years for treatment of HR-positive,
HER2-negative breast cancer.

Mirdametinib is the second drug to be approved for
this indication in the US and the first to be approved
for use in adults. The oral kinase inhibitor selumetinib
(Koselugo) was approved in 2020 for use in patients
2-17 years old.

PIK3CA-related overgrowth spectrum (PROS) who
require systemic treatment. Alpelisib is the first
drug to be approved in the US for this indication.
It was also approved in 2019 as Piqray for use in
combination with fulvestrant for treatment of certain
types of breast cancer.

treatment of metastatic non-small cell lung cancer
(NSCLC) in adults whose tumors have a mutation that
leads to mesenchymal-epithelial transition (MET)
exon 14 skipping. MET exon 14 skipping mutations
occur in 3-4% of NSCLC cases. The drug received
accelerated approval for the same indication in 2020
based on initial overall response rates and duration
of response.
