In Brief: Intensive Glucose Lowering in Type 2 Diabetes

at lower risk has
not been established.
1. The ACCORD Study Group. Effects of intensive glucose lowering ...
The goal for drug therapy of type 2 diabetes is achieving and maintaining a near-normal glycated hemoglobin (HbA1C) concentration without inducing hypoglycemia; the target has generally been an HbA1C of 6.5-7.0% or lower. Whether treating to this level prevents macrovascular (cardiovascular) events has been unclear. Now, 2 large randomized, double-blind trials in patients with long-standing diabetes and at high risk for cardiovascular disease have found no decrease in macrovascular events with intensive glucose control. The ACCORD trial in about 10,000 patients found that patients intensively...

The FDA has approved the use of the phosphodiesterase-5 (PDE-5) inhibitor tadalafil (Cialis – Lilly) for
treatment of signs and symptoms of benign prostatic
hyperplasia (BPH) in men with or without erectile dysfunction.
Tadalafil is approved for use under another brand
name (Adcirca) for treatment of pulmonary arterial hypertension.
The other PDE-5 inhibitors available in the US
for treatment of erectile dysfunction (sildenafil [Viagra];
vardenafil [Levitra]) have also been reported to be effective
for treatment of BPH signs and symptoms, but have
not been approved for this...

Transcatheter aortic-valve replacement (TAVR), a procedure
in which a prosthetic valve is inserted into the
aortic annulus by a catheter, has been approved by the
FDA as an alternative to surgical aortic-valve replacement
for patients with severe symptomatic aortic

The FDA has approved Quartette (Teva), an extended-cycle
combination oral contraceptive containing increasing
doses of the estrogen ethinyl estradiol (EE)
combined with the progestin levonorgestrel (LNG). The
rationale is that a gradual increase in the EE dose may
reduce unscheduled bleeding or spotting, a common
adverse effect of extended-cycle oral contraceptives.

Golodirsen (Vyondys 53 — Sarepta), an antisense
oligonucleotide, has received accelerated approval
from the FDA for treatment of Duchenne muscular
dystrophy (DMD) in the ~8% of patients who have
mutations of the dystrophin gene that are amenable
to exon 53 skipping. It is the first drug to be approved
for this indication and the third to be approved for
treatment of DMD; the antisense oligonucleotide
eteplirsen (Exondys 51) and the oral corticosteroid
deflazacort (Emflaza) were approved earlier.

from the FDA for treatment of Duchenne muscular
dystrophy (DMD) in patients who have mutations
of the dystrophin gene that are amenable to exon
45 skipping (DMD-45), which occur in ~8% of DMD
cases. Casimersen is the first drug to be approved
for this indication and the fifth to be approved for
treatment of DMD; the IV antisense oligonucleotides
