Delandistrogene moxeparvovec-rokl (Elevidys –
Sarepta), an adeno-associated virus (AAV) vector-based
gene therapy, has received accelerated
approval from the FDA for treatment of ambulatory
children 4-5 years old with Duchenne muscular
dystrophy (DMD) who have a confirmed mutation in
the dystrophin gene. It is the first gene therapy to be
approved in the US for treatment of DMD. Accelerated
approval was based on expression of microdystrophin
in skeletal muscle, a surrogate endpoint.

Rituximab (Rituxan - Genentech), an anti-CD20 monoclonal antibody already marketed for treatment of B-cell non-Hodgkin's lymphoma, is now FDA-approved for use concurrently with methotrexate to treat moderate to severe rheumatoid arthritis in patients who have had an inadequate response to one or more tumor necrosis factor (TNF) inhibitors. Rituximab selectively depletes CD20+ B cells, which apparently play a role in the autoimmune response and in the chronic synovitis associated with rheumatoid arthritis.

Coagulation factor Xa (recombinant), inactivated-zhzo
(andexanet alfa; Andexxa – Portola) has received
accelerated approval from the FDA for urgent reversal
of the anticoagulant effect of the direct factor Xa
inhibitors apixaban (Eliquis) and rivaroxaban (Xarelto).
Andexanet alfa is the second antidote for a direct oral
anticoagulant to become available in the US, and the
fi rst for factor Xa inhibitors. Idarucizumab (Praxbind)
was approved in 2015 for reversal of the anticoagulant
effect of the direct thrombin inhibitor dabigatran
etexilate (Pradaxa). Andexanet alfa has not...

autotemcel (Lyfgenia – Bluebird Bio) – have been
approved by the FDA for treatment of sickle cell
disease in patients ≥12 years old with recurrent vaso-occlusive
crises. They are the first gene therapies to
be approved in the US for use in sickle cell disease;
Casgevy is the first treatment to be approved in the
US that uses CRISPR/Cas9 gene-editing technology.

are genetically modified to express chimeric antigen
receptors (CAR) and then reinfused. Kymriah is the
first CAR T-cell immunotherapy to become available in
the US. A CAR T-cell product for B-cell non-Hodgkin's
